Estudio polarográfico del complejo de inclusión progesterona-ciclodextrina

En la presente tesis se estudió el comportamiento electroquímico del complejo de inclusión progesterona-ciclodextrina. Para esto se utilizó β-ciclodextrina (β-CD) y el derivado sulfobutil-β-ciclodextrina (SB-βCD). Este estudio electroquímico fue realizado usando la técnica de polarografía de pulso diferencial (PPD), donde se observó una disminución de la corriente al agregar ciclodextrinas (CDs), esta disminución se debe a que el complejo de inclusión presenta un menor coeficiente de difusión comparado con la molécula libre. Se determinó la constante de asociación para el sistema progesterona-βCD a distintos pH 2,0, 4,0 y 7,5 encontrándose valores de 32, 102 y 850 M-1 respectivamente, lo que muestra una relación entre la formación del complejo y el pH. Se estudió el efecto de la agitación en la formación del complejo a distintos tiempos comparándose con soluciones en reposo. Esto también fue realizado a dos pHs 4,0 y 7,5, donde se observó que, a pH 4,0 la agitación es desfavorable a la formación del complejo; y que a pH 7,5 no tiene gran influencia en la formación del complejo. In the present thesis, the electrochemical behaviour of progesterone- cyclodextrin inclusion complexes was studied. β-cyclodextrin (β-CD) and sulfobutylether-β-cyclodextrin (SBEβCD) were analysed. This study was carried out by differential pulse polarography (DPP) . A decrease of the peak current was observed when cyclodextrins (CDs) were added,. This decrease is explained due to the change of diffusion coefficient. Association constants of progesterone-CD system at different pH 2,0, 4,0 y 7,5 were determined. Constants values of 32, 102 and 850 M-1, respectively were found. These values show a relation between the formation of the complex and pH. An study of the effect of the agitation in the formation of the complex at different times comparing with solutions in rest was performed. This also was carried out at both pH 4.0 and pH 7.5, where it is observed that the agitation is unfavorable to the formation of the complex at pH 4.0 and there is not a great influence in the formation of the complex at pH 7.5.
