Evaluación de la pureza enantiomérica de levofloxacina y su cuantificación en formas farmacéuticas por electroforesis capilar zonal modificada con Sulfobutil- eter-β-ciclodextrina como selector quiral

En este trabajo se presenta el desarrollo y validación de un método simple y rápido para la resolución quiral del racemato de ofloxacina por electroforesis capilar y su aplicación a la industria farmacéutica. Los mejores resultados fueron obtenidos en un sistema electroforético constituído por un buffer de fosfato, 75 mM, pH 2,5 con 0.9 mM de sulfobutil eter-β-ciclodextrina (ciclodextrina aniónica). El voltaje aplicado fue de 13 kV, la detección a 280 nm, la temperatura 25 °C y la inyección hidrostática (10 cm de altura) durante 3s. La influencia de diferentes variables analíticas como naturaleza, concentración y pH del electrolito de corrida, tiempo de inyección y dimensiones del capilar fueron evaluados durante la optimización del método. La validación comprendió la determinación de la linealidad, precisión, límite de detección y cuantificación y exactitud. La aplicabilidad del método fue demostrada en el control de pureza enantiomérica de la levofloxacina y en el análisis de sus formas farmacéuticas como comprimidos, inyectable y gotas óticas.
In this work an optimized, validated, simple and rapid method for the enantiomeric resolution of the racemic form of ofloxacine by capillary electrophoresis is presented. The method was applied to evaluate the chiral impurity profiling of levofloxacine and the quantitation of this drug in pharmaceuticals. The thorough optimization allowed to obtain the best results using an electrophoretic system containing sulfobutyl-ether-3-β-cyclodextrin 0.9 mM in a 75 mM phosphate buffer at pH 2.5. The voltage applied was 13 kV, the detection was set at 280 nm, temperature at 25 °C and the sample was hydrostatically introduced (10 cm height) for 3 s. The effect of different parameters such as nature and concentration and pH of the running electrolyte, injection time and capillary dimensions was evaluated during the optimization of the method. For the method validation the analytical parameters included were linearity, precision, limit of detection and quantitation, accuracy and robutness. The applicability of the method was demonstrated both by evaluating the purity profiling of the levofloxacine and by quantifying this drug in pharmaceutical products.
