Desarrollo de granulados de Fluconazol obtenidos en lecho fluido para producción de cápsulas y tabletas

Fluconazol es un fármaco que presenta baja fluidez, por tanto la finalidad de la presente investigación fue mejorar sus características reológicas, mediante el método de granulación en lecho fluido. Fueron desarrollados ensayos utilizando aglutinantes, tipo almidón modificado y hidroxipropilmetilcelulosa (HPMC). Los granulados obtenidos fueron evaluados en cuanto a características de: velocidad de flujo, ángulo de reposo e índice de compresibilidad, observándose una mejora significativa en las características de fluidez, en la formulación con HPMC. Estos granulados fueron utilizados en la formulación de cápsulas de gelatina y tabletas por compresión directa. Las cápsulas obtenidas se evaluaron en: peso promedio y perfil de disolución y las tabletas se evaluaron en: peso promedio, dureza, friabilidad y perfil de disolución. El perfil y eficiencia de las disoluciones de las formulaciones desarrolladas fueron comparados con el perfil del medicamento de referencia. Las tabletas producidas con ambos granulados mostraron mejor desempeño, siendo aquellas producidas con HPMC (10%), las que se mostraron significativamente superiores en ambas formas farmacéuticas, observados en el perfil y eficiencia de disolución. Fluconazol is a drug that present low fluidity, therefore the purpose of the present research was to improve its reologics characteristics, by means of the fluidized bed granulation. Tests using modified starch and hydroxypropylmethylcellulose (HPMC) were development. The granulated obtained was evaluated based on the following characteristics: powder flowability, angle of repose and compactibility index, being observed improvements significant in the fluidity characteristics, in the formulation with HPMC. These granulates were used in the formulation of capsules of gelatin and tablets by direct compression. The obtained capsules were evaluated in weight average and profile of dissolution and the tablets were evaluated in weight average, hardness, friability and profile of dissolution. The profile and efficiency of the dissolutions of the developed formulations were compared with the profile of the reference medicine. The tablets obtained with both granulated showed better performance, being those produced with HPMC (10%), those that were significantly superior in both pharmaceutical forms, observed in profile and efficiency dissolution.
