Regulatory science governing monoclonal antibody development has undergone a profound transformation over the past decade, shifting from empiricism-driven paradigms toward evidence-efficient, mechanism-based frameworks grounded in analytical rigor and human-relevant biology. This evolution did not originate within innovator drug development in isolation but emerged incrementally through regulatory experience accumulated during biosimilar monoclonal antibody approvals. Across multiple jurisdictions, extensive post-approval evidence demonstrated that routine animal toxicology studies and large comparative clinical efficacy trials rarely contributed decision-relevant information once comprehensive analytical similarity and pharmacokinetic equivalence were established. These observations catalyzed a reassessment of long-standing regulatory assumptions. They culminated in legislative and policy changes that removed the presumption of animal testing and reframed clinical studies as conditional tools to be used only when residual scientific uncertainty persists. As regulatory confidence increased, principles validated in biosimilar development were generalized to novel monoclonal antibodies, resulting in streamlined nonclinical safety expectations and greater reliance on analytical characterization, mechanistic understanding of target biology, and clinical pharmacology. This review synthesizes regulatory guidance and empirical experience with biosimilars. It advances in analytical science, new approach methodologies, and model-informed development to demonstrate that biosimilars function as the regulatory proof-of-concept enabling convergence across innovator and biosimilar pathways. The resulting unified framework emphasizes proportionality, prioritizes evidence predictive of human outcomes, and minimizes redundant testing that adds cost, ethical burden, and limited scientific value. The implications of this regulatory convergence extend beyond efficiency, influencing innovation strategy, global access to biologic therapies, and the sustainability of healthcare systems. Together, these developments mark a decisive shift toward a coherent, evidence-efficient regulatory paradigm for monoclonal antibody development.
