Se realizaron estudios inmunohistoquímicos en muestras fijadas en formol e incluidas en parafina de íleon de 16 bovinos con signos clínicos y lesiones compatibles con paratuberculosis. Los resultados obtenidos en la detección del Mycobacterium paratuberculosis (Mp) mediante las técnicas de Peroxidasa-Antiperoxidasa (PAP), Avidina-Biotina-Peroxidasa (ABC), Streptavidina-Biotina Marcada (LSAB) y de Plata-Inmuno-Oro (IGSS), utilizando un antisuero policlonal específico Mp cepa 2E, se compararon entre sí y con los obtenidos mediante la coloración de Ziehl-Neelsen. El Mp fue detectado, mediante las técnicas de inmunohistoquímica, en 14 de los 16 animales. Los métodos inmunohistoquímicos resultaron los más efectivos para poner en evidencia escasas cantidades de bacilos. La técnica IGSS resultó ser la de mayor sensibilidad aunque es de realización más compleja. Mediante el uso de las lectinas biotiniladas Dolichus biflorus (DBA), Glycine maximus (SBA), Ulex europaeus-l (UEA-I), Triticum vulgaris (WGA), Arachis hypogaea (PNA), Concanavalia ensiformis (Con A), Griffonia simplicifolia / (GS I) y Ricinus communis-l (RCA-I) se demostraron diferencias cuando se compararon los patrones de afinidad de los animales afectados y de 5 animales utilizados como control. Estas diferencias demostrarían deficiencias de los enterocitos y de las células caliciformes en la producción de mucinas. También se realizaron estudios ultraestructurales en 6 casos con lesiones severas de la enfermedad. Se observaron cambios degenerativos en los enterocitos, bacterias libres en el citoplasma y también en los fagosomas de los macrófagos. En los fagolisososmas se constataron microorganismos intactos y fragmentados. Immunohistochemical studies were performed in formalin-fixed, paraffinembedded ileum samples of 16 bovines with clinical signs and lesions compatible with paratuberculosis. In order to detect Mycobacterium paratuberculosis (Mp) the following immunohistochemical methods were employed: Peroxidase- Antiperoxidase (PAP), Avidina-Biotina-Complex (ABC), Labelled Streptavidin- Biotine (LSAB) and Immuno-Gold-Silver-Stain (IGSS) using a commercial hyperimmune serum against Mp strain 2E. The results were compared to each other and with those obtained by means of Ziehl Neelsen (ZN). The Mp was detected in 14 of the 16 animals. The immunohistochemical techniques were more effective to detect small number of bacilli. The IGSS provided higher sensitivity although it is a more complex to be carried out. The lectin binding pattern was also studied in 16 affected animals as well as in 5 normal animals. The following biotinylated lectines were applied: Dolichus biflorus (DBA), Glycine maxim us (SBA), Ulex europaeus-\ (UEA-I), Triticum vulgaris (WGA), Arachis hypogaea (PNA), Concanavalia ensiformis (Con-A), Griffonia simplicifolia-\ (GS I) and Ricinus communis-\ (RCA-I). The modifications observed would demonstrate deficiencies of the enterocytes and the goblet cells in the production of glycoconjugates. Ultrastructural studies were also carried out in advanced cases of the disease. Degenerative changes were seen in the epithelial cells free bacilli in both the cytoplasm and the phagosomas of the macrophages were also found. In adition, intact and fragmented microorganisms were observed into the phagolysosomes.
