Bovine herpesvirus 1 (BHV-1) y Bovine herpesvirus 5 (BHV-5) pertenecen a la familia Her- pesviridae, subfamilia Alphaherpesvirinae, género Varicellovirus [1]. BHV-1 presenta una distribución mundial; produce conjuntivitis, rinotraqueítis infecciosa bovina (IBR), vulvovaginitis pustular infecciosa (IPV) y balanopostitis (IBP). También se asocia a enteritis, aborto y muerte en neonatos. BHV-5 afecta a animales de entre 5 y 24 meses de edad, produciendo cuadros de meningoencefalitis generalmente mortales. En nuestro país los trabajos serológicos realizados han indicado prevalencias mayores al 40 % en todas las regiones investigadas. BHV-5 fue aislado por primera vez en 1982; aún no se conocen los aspectos epidemiológicos relacionados con la infección por este virus. En esta tesis se realizaron inoculaciones experimentales en bovinos. Estas experiencias permitieron reproducir los síntomas clínicos característicos de BHV-1 y BHV-5, obtener especímenes para el diagnóstico de estos agentes y además, multiplicar el virus en su huésped natural. Por otro lado, se diseñó una PCR-M que utiliza primers específicos para cada especie viral, de manera que puede ser utilizada como herramienta diagnóstica diferencial entre ambas entidades. Se evaluaron diferentes aspectos inherentes a la capacidad diagnóstica de la técnica tales como su sensibilidad y especificidad, y además, su capacidad diagnóstica en diferentes muestras clínicas (hisopados nasales, órganos, semen). Además se analizaron 20 aislamientos de campo que habían sido previamente caracterizados mediante el uso de anticuerpos monoclonales. En todos los casos los resultados demostraron que la PCR-M desarrollada en este trabajo de tesis, puede ser utilizada como herramienta diagnóstica diferencial entre BHV-1 y BHV-5. Este trabajo representa un nuevo aporte al estudio de las enfermedades causadas por estos virus, ya que hasta el momento BHV-1 y BHV-5 no podían ser diferenciados por los métodos de diagnóstico virológico clásicos, y tampoco existía una técnica molecular diferencial que resulte adecuada para su uso en los laboratorios de diagnóstico. Bovine herpesvirus 1 (BHV-1) and Bovine herpesvirus 5 (BHV-5) are members of the genus Varicellovirus, which is classified in the family Herpesviridae, subfamily Alphaherpesvirinae [1]. BHV-1 presents a worldwide distribution. Infections of cattle with this agent are related to conjunctivitis, infectious bovine rhinotracheitis (IBR), infectious pustular vulvovaginitis (IPV) and balanoposthitis (IBP). It can also cause enteritis, abortion and death in newborn calves. BHV-5 affects 5 to 24 months old animals, producing fatal outbreaks of meningoencefalitis. Previous works have shown that more than 40% of cattle are seropositive against BHV-1 in Argentina. In Argentina, BHV-5 was first isolated in 1982. Epidemiological studies of BHV-5 are scarce. In this thesis, experimental inoculations of cattle with BHV-1 and BHV-5 are described. Clinical presentation of both agents were reproduced during these experiences, which also provided specimens to be used in the PCR-method check out. The controlled trials also showed that these viruses can be multiplied in its natural host without major difficulties. A PCR able to detect and differentiate BHV-1 and BHV-5 was developed. The technique uses specific primers against each agent. Both primer pairs were combined in a Multiplex-PCR (PCR-M). PCR-M was evaluated by the analyses of its sensitivity and specificity. The ability of the method to detect virus in different specimens (nasal swabs, organs, semen) was evaluated. Furthermore, twenty field isolates previously characterized by monoclonal antibodies were submited to viral detection and typing by PCR-M. All the analyses showed the PCR-M would be used as a diagnostic tool against BHV-1 and BHV-5. Both species were always succesfully differentiated by the PCR-M. The present work constitutes an improvement in knowledge on bovine herpesviruses, since classical or molecular virological methods for typing these agents were not available before.
