Summary

The diffusional resistances that the drug molecules have to overcome during the course of penetration across the skin tissue and the subsequent uptake by the capillary network for transport to the general circulation are described. The mean diffusional barrier has been found to resid in the stratum corneum cell layers. If the drug is delivered to the skin via a transdermal therapeutic system (T.T.S.) at a lower releasing rate than the rate of percutaneous absorption, the process of drug release will play the rate-controlling role to the blood levels of drugs. The zero, first and 112 order rate constant of the drug delivery process from a T.T.S. are considered. Also, the factors that affect the rate permeation of drug through the skin are analyzed.
