La Fusariosis de la espiga de trigo (FET) es una compleja enfermedad fúngica que afecta tanto al rendimiento y calidad de la producción granaria como a la salud por acción de la micotoxina asociada. Diversas especies de Fusarium pueden relacionarse con la FET. La distribución y predominancia de estos patógenos están determinadas en gran parte por factores climáticos como la temperatura y la humedad. En Argentina, el agente fúngico dominante asociado a la FET es Fusarium graminearum (Schwabe) anamorfo de Gibberella zeae (Schw.) Petch. La enfermedad es el resultado de la acción individual o combinada de diferentes mecanismos de patogénesis, como la producción y liberación de enzimas extracelulares que degradan los principales constituyentes de la pared celular y permiten la colonización de los tejidos. Una vez establecida la infección se produce la liberación de micotoxinas que interfieren con el metabolismo o que afectan la integridad estructural de la célula hospedera o bien sus procesos fisiológicos, lo cual da como resultado pérdida en el rendimiento y calidad de las cosechas. La investigación se basó en el aislamiento e identificación de aislados de Fusarium spp. asociados con FET a partir de muestras de trigo provenientes de 15 localidades distribuidas en las provincias de Buenos Aires, Entre Ríos, Santa Fe y Córdoba durante las campañas trigueras 2006 y 2007. Se analizó la diversidad genética entre aislamientos de F. graminearum con marcadores moleculares ISSR y RFLP, así como la diversidad química y estructural mediante espectroscopía vibracional FT-IR. Determinada la distancia genética entre aislamientos, se analizó la agresividad de los mismos sobre trigo en invernáculo a partir del análisis de diversas variables patométricas. Además, se detectaron y caracterizaron actividades enzimáticas relacionadas con el proceso de colonización y daño sobre los granos, las cuales serían responsables de cambios estructurales y de composición de los mismos. Se seleccionó un aislamiento de F. graminearum con elevada actividad enzimática poligalacturonasa, como estimativo de agresividad, para evaluar resistencia a la penetración del patógeno en la espiga. La patogenicidad se evaluó sobre diversos genotipos de trigo a campo bajo condiciones de temperatura y humedad parcialmente controladas. Este tipo de evaluación permite identificar nuevas fuentes de resistencia. Fusarium head blight (FHB) is a complex fungal disease that affects both the yield and quality of grain production, as health by action of the mycotoxin associated. Several species of Fusarium can relate to the FHB. The distribution and prevalence of these pathogens is largely determined by climatic factors like temperature and humidity. In Argentina, the dominant fungal agent associated with FHB is F. graminearum (Schwabe) anamorph of Gibberella zeae (Schw.) Petch. The disease is the result of individual or combined action of different mechanisms of pathogenesis, as the production and release of extracellular enzymes that degrade the major constituents of the cell wall and allows colonization of tissues. Once established the infection, the pathogen produces mycotoxins that interfere with the metabolism or affect the structural integrity of the host cell or physiological processes, resulting in loss in yield and quality of crops. The research was based on isolation and identification of isolates of Fusarium spp. associated with FHB of wheat obtained from samples of wheat grains from Pampas region from 15 locations distributed in Buenos Aires, Entre Ríos, Santa Fe and Córboba provinces during 2006 and 2007 wheat growing seasons. The genetic diversity among isolates of F. graminearum was analyzed by ISSR and RFLP molecular markers, as well as their chemical and structural diversity by FT-IR vibrational spectroscopy. After determining the genetic distance among the isolates, their aggressiveness on wheat in greenhouse was analyzed from the analysis of pathomerics variables. Also, enzymatic activities related to the process of colonization and damage to the grains were detected and characterized, which could be responsible for structural and constitutional changes. An isolate of F. graminearum with high polygalacturonase activity as aggressiveness criterion was selected and used to evaluate the resistance to penetration of the pathogen into the spike. Pathogenicity was evaluated on various wheat genotypes in test to field with partial control of temperature and humidity. This type of evaluation allows identifying new sources of resistance.
